Cells synthesize glutathione through two ATP-dependent steps: glutamate-cysteine ligase joins glutamate and cysteine, and glutathione synthetase adds glycine to form the complete tripeptide. Breakdown occurs through gamma-glutamyl transpeptidase and subsequent peptidase reactions, forming the gamma-glutamyl cycle. Within cells, glutathione also forms a disulfide-linked dimer called GSSG when two GSH molecules react. The balance between GSH and GSSG is widely used as an indicator of oxidative conditions, although the ratio can vary by compartment and tissue.
Glutathione supports several cellular processes beyond direct antioxidant action. It serves as a cofactor for glutathione peroxidase and glutathione S-transferase enzymes, which reduce peroxides and conjugate electrophiles, respectively. The molecule also acts as a reservoir of cysteine, an amino acid that can limit protein synthesis and redox signaling. In human nutrition, oral glutathione is sold as a supplement, but how much intact glutathione reaches tissues after ingestion remains an active research question. Clinical claims about supplementation are not uniformly supported by controlled trials.
Storage recommendations for glutathione reagents usually specify a cool, dry, dark environment because the thiol oxidizes in air and light. Solid material is often kept desiccated at low temperature, while solutions are prepared fresh or stored frozen in aliquots. Repeated freeze-thaw cycles can accelerate degradation, and metal ions can catalyze oxidation. Quality control may include purity assays, water content, and identity confirmation. Stability limits are method-specific, so a stated shelf life applies only to defined conditions and packaging.
Laboratory measurement of glutathione requires attention to oxidation before analysis. Blood, tissue, or cell samples can lose reduced glutathione as it converts to GSSG or forms mixed disulfides with proteins. Acid extraction, rapid freezing, and thiol-blocking reagents are common strategies to preserve the original distribution. Reported concentrations therefore depend on collection protocol, extraction method, and the time between sampling and analysis. Comparisons across studies are most reliable when these pre-analytical variables are described.
Common analytical approaches include enzymatic recycling assays, high-performance liquid chromatography, and mass spectrometry. The enzymatic recycling assay uses glutathione reductase and a colorimetric or fluorometric reagent to amplify signal, which gives good sensitivity for total glutathione. Chromatographic methods can separate GSH from GSSG and related thiols, while mass spectrometry offers structural confirmation and multiplexing. Each approach has different requirements for calibration, internal standards, and validation. No single method captures every form of glutathione in every matrix.

Synthesis occurs in two ATP-dependent steps: glutamate-cysteine ligase joins glutamate and cysteine to form gamma-glutamylcysteine, and glutathione synthetase adds glycine to complete the tripeptide. The pathway is feedback-inhibited by GSH and limited by cysteine availability, so cysteine supply often constrains production. Once formed, GSH participates in redox buffering, xenobiotic conjugation, and protein glutathionylation. Glutathione peroxidase uses GSH to reduce hydrogen peroxide and lipid peroxides, yielding GSSG, while glutathione reductase regenerates GSH using NADPH. Glutathione S-transferases conjugate electrophiles to GSH, supporting detoxification and excretion.
Because GSH is central to redox balance, its status is studied in aging, liver disease, neurodegenerative conditions, and metabolic disorders. Observational studies often report lower GSH or higher GSSG in affected tissues, but such associations do not establish that raising glutathione changes disease outcomes. Oral glutathione is digested into amino acids, and whether intact absorption occurs remains debated; precursors such as N-acetylcysteine and cysteine donors are also investigated. Regulatory agencies generally treat glutathione as a dietary supplement, not an approved drug, and clinical claims require evidence from controlled trials.
Measuring glutathione requires attention to sample preparation because the molecule oxidizes readily. Blood, tissue, and cell samples are often treated with acid to precipitate proteins and stabilize the thiol. Without such steps, GSH can convert to GSSG or form mixed disulfides during storage. Analytical methods include spectrophotometric assays, high-performance liquid chromatography, and mass spectrometry. Each approach has different sensitivity, specificity, and susceptibility to interference from related compounds in complex matrices.
For solid glutathione, storage conditions affect shelf life. The reduced form is typically kept cool, dry, and protected from air and light. Moisture can promote oxidation, while elevated temperatures accelerate degradation. Suppliers often specify storage at or below freezing, sometimes under inert gas. Solutions are less stable than powders and may require preparation shortly before use. Buffers and chelating agents can slow oxidation, but they do not eliminate it. Published stability data vary with matrix, pH, and container.
== Early life and education == Namandjé Bumpus was born in Philadelphia and raised in western Massachusetts. She became interested in chemistry at a young age, even writing to the American Chemical Society while still in elementary school to ask about the kind of careers chemists can have. She earned a B.A. in Biology from Occidental College, in Los Angeles, California, in 2003. At Occidental, she was introduced to research experiences in ecology, then she ventured into pharmacology through Charles Ross Summer Research Fellowship at the University of Michigan, during which she was mentored by Dr. Richard R. Neubig. She enjoyed the experience so much that she decided to return to the University of Michigan after graduating from Occidental College in order to pursue a PhD in pharmacology. She earned her Ph.D. in pharmacology from the University of Michigan Medical School in 2007. Her thesis research, and much of her later work, examined how drugs are processed by cytochrome P450 enzymes, (CYPs) a family of heme-containing monooxygenases, that often help make drugs more soluble, aiding with drug clearance. Bumpus performed her thesis research in the laboratory of Dr. Paul F. Hollenberg, investigating how a naturally occurring mutation in CYP2B6 affects its ability to be inactivated by compounds known the inactivate the wild-type CYP2B6. She also looked into how naturally occurring variants could impact how patients cleared the antidepressant Bupropion, and the antiviral Efavirenz.
In biochemistry, a kinase (, EC 2.7.-.-) is a protein enzyme that catalyzes the transfer of phosphate groups to substrates. This process is known as phosphorylation. Typically ATP is the phosphate donor. Kinases are pervasive, the human genome codes for about 500 of these enzymes.
It is important for MADD patients to maintain strength and fitness without exercising or working to exhaustion. Learning this balance may be more difficult than normally, as muscle pain and fatigue may be perceived differently from normal individuals. Symptomatic relief from the effects of MADD may sometimes be achieved by administering ribose orally at a dose of approximately 10 grams per 100 pounds (0.2 g/kg) of body weight per day, and exercise modulation as appropriate. Taken hourly, ribose provides a direct but limited source of energy for the cells. Patients with myoadenylate deaminase deficiency do not retain ribose during heavy exercise, so supplementation may be required to rebuild levels of ATP. Creatine monohydrate could also be helpful for AMPD patients, as it provides an alternative source of energy for anaerobic muscle tissue and was found to be helpful in the treatment of other, unrelated muscular myopathies. The ambassador from the Netherlands explained his negative vote as being due to the reference in the preamble to the resolution "to potential harmful effects of the use of depleted uranium munitions on human health and the environment [which] cannot, in our view, be supported by conclusive scientific studies conducted by relevant international organizations." None of the other permanent members of the United Nations Security Council supported the resolution as China was absent for the vote, Russia abstained and the United States voted against the resolution. In September 2008, and in response to the 2007 General Assembly resolution, the UN Secretary General published the views of 15 states alongside those of the International Atomic Energy Agency (IAEA) and World Health Organization (WHO). The IAEA and WHO evidence differed little from previous statements on the issue. The report was largely split between states concerned about depleted uranium's use, such as Finland, Cuba, Japan, Serbia, Argentina, and predominantly NATO members, who do not consider the use of depleted uranium munitions problematic. In December 2008, 141 states supported a resolution requesting that three UN agencies: United Nations Environment Programme (UNEP), WHO and IAEA update their research on the impact of uranium munitions by late 2010—to coincide with the General Assembly's 65th Session, four voted against, 34 abstained and 13 were absent. As before Britain and France voted against the resolution.
== Toxicity == Highly charged and heavy, alpha particles lose their several MeV of energy within a small volume of material, along with a very short mean free path. This increases the chance of double-strand breaks to the DNA in cases of internal contamination, when ingested, inhaled, injected or introduced through the skin. Otherwise, touching an alpha source is typically not harmful, as alpha particles are effectively shielded by a few centimeters of air, a piece of paper, or the thin layer of dead skin cells that make up the epidermis; however, the decay chain of many alpha emitting isotopes, for example Radium 226, contain daughter nuclei that undergo beta- and/or gamma-decay. Relative biological effectiveness (RBE) quantifies the ability of radiation to cause certain biological effects, notably either cancer or cell-death, for equivalent radiation exposure. Alpha radiation has a high linear energy transfer (LET) coefficient, which is about one ionization of a molecule/atom for every angstrom of travel by the alpha particle. The RBE has been set at the value of 20 for alpha radiation by various government regulations. The RBE is set at 10 for neutron irradiation, and at 1 for beta radiation and ionizing photons. However, the recoil of the parent nucleus (alpha recoil) gives it a significant amount of energy, which also causes ionization damage (see ionizing radiation). This energy is roughly the weight of the alpha (4 Da) divided by the weight of the parent (typically about 200 Da) times the total energy of the alpha.

The elections, while a landmark of democratic transformation, were also shaped significantly by external influences particularly from West Germany. Writer Michael Schneider was among those who criticised what he viewed as overwhelming interference by West German politicians and institutions. In his work, he described the campaign environment as one saturated with West German personalities, party activists and messaging. West German political parties, especially its branches of the Christian Democratic Union (CDU) and Social Democratic Party (SPD), sent campaigners and resources across the border to bolster their East German affiliates. According to Schneider, taxpayer funds from West Germany were used to support this mobilisation, raising questions about the balance of influence and the fairness of the electoral playing field. Civil rights activist and co-founder of the New Forum, Jens Reich, echoed these concerns from the perspective of the East German opposition. Speaking two decades after the vote, he reflected that the entire West German political apparatus had been transplanted into the East, overwhelming grassroots democratic movements that had played a key role in the Peaceful Revolution. In his words, "The Bonn hippopotamus came in such a mass that you were simply helpless." Reich observed that the election campaign bore little resemblance to a homegrown democratic process and instead felt like a western-style election exported wholesale into a fragile new democracy.
Trypanothione is an unusual form of glutathione containing two molecules of glutathione joined by a spermidine (polyamine) linker. It is found in parasitic protozoa such as leishmania and trypanosomes. These protozoal parasites are the cause of leishmaniasis, sleeping sickness and Chagas' disease. Trypanothione was discovered by Alan Fairlamb. Its structure was proven by chemical synthesis. It is present mainly in the Kinetoplastida but can be found in other parasitic protozoa such as Entamoeba histolytica. Since this thiol is absent from humans and is essential for the survival of the parasites, the enzymes that make and use this molecule are targets for the development of new drugs to treat these diseases. Trypanothione-dependent enzymes include reductases, peroxidases, glyoxalases and transferases. Trypanothione-disulfide reductase (TryR) was the first trypanothione-dependent enzyme to be discovered (EC 1.8.1.12). It is an NADPH-dependent flavoenzyme that reduces trypanothione disulfide. TryR is essential for survival of these parasites both in vitro and in the human host. A major function of trypanothione is in the defence against oxidative stress. Here, trypanothione-dependent enzymes such as tryparedoxin peroxidase (TryP) reduce peroxides using electrons donated either directly from trypanothione, or via the redox intermediate tryparedoxin (TryX). Trypanothione-dependent hydrogen peroxide metabolism is particularly important in these organisms because they lack catalase.
Panini sticker albums were again announced for the 2026 World Cup. Due to the expansion of the tournament, each pack now contained seven stickers as opposed to the usual five. In the United States and Canada, a set of twelve stickers was reserved for stickers that could only be obtained from Coca-Cola bottles, with each bottle containing a sticker printed inside the label. In May 2026, it was announced that the 2026 World Cup would be Panini's second-to-last tournament with a sticker album, after FIFA announced that Fanatics (via their Topps brand) would distribute collectables for FIFA tournaments from 2031. In conjunction with the tournament, the Lego Group released a series of officially licensed FIFA World Cup 2026-themed construction sets. The collection included models of the FIFA World Cup Trophy, the official tournament emblem, a brick-built Adidas Trionda soccer ball containing a miniature stadium, and player-focused sets based on soccer players Cristiano Ronaldo, Lionel Messi, Kylian Mbappé, and Vinícius Júnior. Several sets featured hidden references to the players' careers, alternative display poses, and commemorative World Cup-themed designs. To promote the product line, Lego organized FIFA World Cup 2026-themed activations in several markets, including a soccer festival and pop-up experience in Singapore. The events featured interactive soccer challenges, soccer-themed Lego builds, displays of player-inspired models, and promotional giveaways tied to the World Cup collection.
The Leidenfrost effect or film boiling is a physical phenomenon in which a liquid, close to a solid surface of another body that is significantly hotter than the liquid's boiling point, produces an insulating vapor layer that keeps the liquid from boiling rapidly. Because of this repulsive force, a droplet hovers over the surface, rather than making physical contact with it. The effect is named after the German doctor Johann Gottlob Leidenfrost, who described it in A Tract About Some Qualities of Common Water. This is most commonly seen when cooking, when drops of water are sprinkled onto a hot pan. If the pan's temperature is at or above the Leidenfrost point, which is approximately 193 °C (379 °F) for water, the water skitters across the pan and takes longer to evaporate than it would take if the water droplets had been sprinkled onto a cooler pan.

It is a tripeptide of glutamic acid, cysteine, and glycine. The linkage between glutamate and cysteine uses the gamma-carboxyl group, which is unusual for peptides.
GSH is the reduced form with a free thiol group. GSSG is the oxidized dimer formed when two GSH molecules join by a disulfide bond.
It is synthesized inside cells and is not classified as an essential dietary nutrient for most people. Dietary and supplemental sources are studied, but direct requirements are not established in the same way as for vitamins.
Pre-analytical handling, extraction chemistry, and detection method all influence reported glutathione values. Oxidation during sample processing can shift the measured GSH/GSSG ratio. Standardized protocols and reference materials help reduce, but do not eliminate, these differences.
